fa شناسایی مولکولی و فیلوژنی گونه‌های جنس Nerita در سواحل صخره‌ای شمال خلیج فارس بيوسيستماتيك آبزيان بيوسيستماتيك آبزيان پژوهشي Research <p><span dir="RTL">در این پژوهش شناسایی مولکولی گونه&shy;های جنس </span><em>Nerita</em><span dir="RTL">، شامل </span><em>N.</em> <em>lognii</em><span dir="RTL">، </span><em>N. albicilla </em><span dir="RTL">و </span><em>N. polita</em><span dir="RTL">، در سواحل شمالی خلیج&shy;فارس به همراه بررسی مورفولوژیک آن&shy;ها انجام شد. این جنس متعلق به خانواده </span>Neritidae<span dir="RTL"> بوده و از گونه&shy;های غالب شکم&shy;پایان سواحل صخره&shy;ای می&shy;باشد. نمونه&shy;برداری در سال&shy;های 92 و 93 در سواحل صخره&shy;ای صورت گرفت. نمونه&shy;ها مورد شناسایی مورفولوژیک قرار گرفتند و سپس مراحل استخراج </span>DNA<span dir="RTL"> (با استفاده از پودر </span>Chelex<span dir="RTL">)، تکثیر قطعه ژنی زیر واحد </span>I<span dir="RTL"> سیتوکروم اکسیداز (</span>COI<span dir="RTL">) و </span>S rRNA<span dir="RTL">16 و با روش سنجر توالی&shy;یابی انجام شد. در مجموع تعداد 6 توالی </span>COI<span dir="RTL"> و 6 توالی </span>S<span dir="RTL">16 متعلق به 3 گونه از جنس </span><em>Nerita</em>&nbsp; <span dir="RTL">به&shy;دست آمد که این توالی&shy;ها برای اولین بار از منطقه شمالی خلیج&shy;فارس گزارش شده است. آنالیزهای فیلوژنی با استفاده از نرم&shy;افزارهای </span>MEGA6<span dir="RTL"> و </span>BEAST<span dir="RTL"> و با رسم درخت&shy;های فیلوژنی </span>Maximum Likelihood<span dir="RTL"> و </span>Bayesian<span dir="RTL"> صورت گرفت. نتایج حاصل از شناسایی مولکولی گونه&shy;های </span><em>N. longii </em><span dir="RTL">و </span><em>N. albicilla</em><span dir="RTL"> با شناسایی سنتی و مورفولوژیک مطابق بود؛ ولی درمورد گونه </span><em>N. polita </em><span dir="RTL">تفاوت&shy;هایی میان آن&shy;ها مشاهده شد که می&shy;تواند نشان دهنده اهمیت مطالعه هم&shy;زمان مولکولی و مورفولوژیک باشد. همچنین&shy; نتایج نشان داد</span> <span dir="RTL">که گونه&shy;های جنس </span><em>Nerita</em><span dir="RTL"> مونوفیلتیک می&shy;باشند.&nbsp;</span></p> <p><em>Nerita</em>s are among the most dominant groups of Gastropoda in the Persian Gulf. There is no previous study in relation to molecular and phylogeny of <em>Nerita</em>s in the study area. The molecular identification of <em>Nerita</em> species have been studied for the first time in the northern rocky coastal zones of Persian Gulf during 2013 and 2014. After morphological identifications of species, DNA extraction, amplifying partial of cytochrome oxidase COI and 16S rRNA and sequencing procedure were done in the laboratory. In this study, 6 COI and 6 16S rRNA sequences, belonging to 3 species, were obtained. Also, phylogeny analyses with drawing phylogeny trees of Maximum Likelihood and Bayesian were done using MEGA6 and BEAST softwares. Morphological and molecular identification results were similar for 2 species and dissimilar for one species. This discrepancy shows that a combination of morphological and molecular studies is more reliable for species identification than either of them solely.
