fa بررسی آرایه‌شناسی گونه‌های جنسAlburnoides در ایران با استفاده از زیر واحد 1 ژن سیتوکروم اکسیداز COI Phylogeny of the members of the genus Alburnoides in Iran using COI gene بيوسيستماتيك آبزيان بيوسيستماتيك آبزيان پژوهشي Research <div style="text-align: justify;">تاکنون 12 گونه از کپورماهیان جنس Alburnoides در ایران بر اساس صفات ریخت‌شناسی توصیف‌شده است. ویژگی‌های کلیدی تخصیص داده ‌شده به این گونه‌ها فاقد کارایی لازم جهت شناسایی آنهاست، بنحوی که با استفاده از این ویژگی‌ها نمی‌توان گونه‌های توصیف‌ شده را از یکدیگر تشخیص داد. از اینرو، این تحقیق به ‌منظور بررسی روابط آرایه‌شناسی و اعتبارسنجی گونه‌های توصیف ‌شده با استفاده از نشانگر مولکولی زیر واحد I ژن سیتوکروم اکسیداز (COI) به اجرا درآمد. برای این منظور تعداد 70 نمونه از اعضاء این جنس از 20 ایستگاه مختلف در 6 حوضه آبریز داخلی نمونه‌برداری شد. جهت تعیین تفاوت‌های ژنتیکی و روابط خویشاوندی، DNA نمونه‌ها به روش فنل-کلروفرم استخراج و ژن سیتوکروم COI طی فرآیند PCR تکثیر و پس از خالص‌سازی توالی‌‌یابی شد. نتایج نشان داد که گونه‌های ایرانی جنس Alburnoides به هشت شاخه تقسیم می‌شوند. بیشترین فاصله ژنتیکی K2P درون گونه‌ای، مابین نمونه‌های گونهA. samiii  و کمترین میزان بترتیب در بین نمونه‌های A. coadi، A. holciki، A. parhami و A. nicolausi محاسبه گردید. همچنین بیشترین فاصله ژنتیکی K2P بین‌گونه‌ای به میزان 56/7 بین دو گونه A. qanati و A. holciki و کمترین میزان به مقدار 33/0 بین دو گونه A. parhami و A. holciki بدست آمد. با توجه به نتایج، به‌‌ دلیل تمایز اندک ژنتیکی بر اساس ژن COI، اعتبار گونه‌های A. parhami،A. coadi  و A. nicolausi تائید نگردید و پیشنهاد می‌‌گردد که بترتیب مترادف گونه‌های A. holciki، A. namaki و A. idignensis در نظر گرفته شوند.</div> <div style="text-align: justify;">Up to now, twelve species of the genus Alburnoides belonged to Cyprinidae have been described from Iranian inland water solely based on the morphological characteristics. The described key characters for these species have no efficiency for their discrimination, and cannot help their precise diagnosis. Therefore, this study was conducted to study the phylogenetic relationship and verification of these species based on COI gene. For this purpose, 70 specimens of Alburnoides species were collected from 20 sampling sites in six Iranian inland water basins. To determination of inter- and intra-species genetic differences, the DNA of sampled specimens were extracted based on Phenol-chloroform method, their COI gene were amplified using thermal cycler and were sequenced after purifications. Based on the results, the members of the Iranian species of the genus Alburnoides were divided into seven linages. Maximum and minimum intra-species genetic distances based on K2P criterion were found between the populations of A. samiii, and A. coadi, A. holciki, A. parhami and A. nicolausi, respectively. Furthermore, maximum and minimum inter-species genetic distances based on K2P criterion were calculated between A. qanati and A. holciki (7.56), and A. parhami and A. holciki (0.33).
