fa شناسایی مولکولی و فیلوژنی گونه Gammarus komareki (Gammaridae) از رودخانه جاجرود با روش بارکدگذاری DNA Molecular and phylogenic identification of Gammarus komareki species (Gammaridae) from the Jajrood River using DNA barcoding method بيوسيستماتيك آبزيان بيوسيستماتيك آبزيان پژوهشي Research گاماروس‌‌ها از انواع سخت‌‌پوستان ناجورپا متعلق به خانواده Gammaridae بوده که نقش و اهمیت زیادی در زنجیره غذائی، پالایش محیط از مواد آلی و حساسیت بالایی نسبت به آلودگی‌های محیطی دارند. بنابراین، شناسایی آنها از اهمیت زیادی برخوردار است. از آنجایی‌‌که شناسایی جنس گاماروس به‌‌ دلیل ویژگی‌‌های ریخت‌‌شناسی شبیه به هم بسیار مشکل می‌‌باشد، هدف مطالعه حاضر شناسایی دقیق مولکولی و بررسی روابط فیلوژنی گونه komareki Gammarusاز رودخانه جاجرود در شرق استان تهران در مقایسه با سایر نقاط ایران و جهان بود. بدین منظور، جمع‌‌آوری و نمونه‌‌برداری با استفاده از تور دستی کوچک از گونه G. komareki رودخانه جاجرود به صورت فصلی از مهر 1392 لغایت تیر 1393 صورت پذیرفت.  به منظور مطالعات مولکولی مقداری بافت از هر نمونه با اسکالپل جدا و در الکل اتیلیک 95 درجه تثبیت گردید. سپس DNA نمونه‌‌ها با استفاده از روش CTAB استخراج و قطعه ژنی زیر واحد یک سیتوکروم اکسیداز میتوکندری (COI) تکثیر و توالی‌‌یابی شد. پس از تکثیر قطعه ژن و توالی‌‌یابی آن، درخت‌‌های تبارشناسی Neighbor Joining وMaximum Likelihood با استفاده از نرم‌‌افزارهای MEGA7 ترسیم گردیدند. نتایج مطالعه حاضر نشان داد، اگرچه نمونه‌‌های موردنظر از نظر ریخت‌‌شناسی متعلق به گونه komareki  Gammarus می‌‌باشند که از سایر مناطق ایران گزارش شده است، با این‌‌حال بررسی‌‌های مولکولی نشان داد نمونه‌‌های مطالعه حاضر با قرار گرفتن در دو کلاد مختلف به دو دسته تقسیم شدند. بنابراین، این احتمال وجود دارد که شاید این نمونه‌‌ها متعلق به گونه‌‌های جداگانه ‌‌باشند. که اثبات این امر نیازمند بررسی‌‌های دقیق‌‌تر با سایر نشانگرهای ژنتیکی می‌‌باشد.   Gammarus (Crustacea: Amphipoda) are Gammaridae family that play an important role in food chain, environmental refining of organic matter and high sensitivity to environmental contamination. Therefore, it is important to identify them. Since the identification of the genus Gammarus is very difficult due to similar morphological features, the aim of the present study was to molecular identification and investigation of the phylogenetic relationships of G. komareki from the Jajrood River, east of Tehran compared to other parts of Iran and the rest of the world. For this purpose, sampling of G. komareki species in Jajrood River was done in season from October 2013 to July 2014. Samples were collected by a small manual tour. For molecular studies, some tissue was isolated from each sample with scalpel and fixed in 95% ethyl alcohol. The DNA of the samples was then extracted using the CTAB method and the gene fragment of a mitochondrial cytochrome oxidase (COI) subunit was amplified and sequenced. After gene fragment amplification and sequencing, Neighbor Joining and Maximum Likelihood trees were plotted using MEGA7 software. The results of the present study showed that although the specimens were morphologically belonging to Gammarus komareki species reported from other parts of Iran, Molecular studies showed that the samples of the present study were divided into two categories by being placed in two different clouds.
